      TORCH感染的早期快速诊断在疾病管理中至关重要,有助于提高诊疗效果,降低医疗风险。利用多重荧光编码技术,建立了一种可同时检测弓形虫、风疹病毒、巨细胞病毒、单纯疱疹病毒特异性抗体IgG的免疫分析法。通过对偶联缓冲液pH值及Goat anti-Human IgG- PE浓度的优化得到最佳检测条件,并对该实验方法的精密度、特异性、剂量-反应曲线及检出限进行性能验证。结果表明,各项目的相对标准偏差(RSD)均小于10%,与多种常见的病原体抗体均无交叉反应,在40 min内能同时完成5个项目的批量检测,且与CLIA方法测试的结果具有良好的相关性。此方法可缩短检验周期、节约检测成本,为临床上快速、灵敏、高通量的TORCH感染筛查提供一种新的途径。
英文摘要:
      Rapid early diagnosis of TORCH infection is crucial in disease control and helps to improve the effectiveness of diagnosis and treatment, and reduce medical risks. An immunoassay for simultaneous detection of IgG specific antibodies against Toxoplasma gondii, rubella virus, cytomegalovirus and herpes simplex virus was established using multiplex fluorescence coding technique. The optimal assay conditions were achieved by optimizing the pH of the coupling buffer and the concentration of Goat anti-Human IgG-PE. The performance of the modified experimental method was verified in terms of precision, specificity, dose-response curve and detection limit. The results showed that the relative standard deviation (RSD) of each item was less than 10%, and there was no cross-reactivity with antibodies to a variety of common pathogens. The batch testing of five items could be completed simultaneously within 40 minutes, and the results were well correlated with those tested by CLIA method. This method can shorten the test cycle and reduce testing cost, providing a new option for rapid, sensitive and high-throughput TORCH infection screening in the clinic.
